Background
DDX39B (DEAD-box helicase 39B), also known as UAP56 or Bat1, is an ATP-dependent RNA helicase belonging to the DEAD-box family that participates in multiple critical steps of RNA metabolism within the nucleus. As an important component of the spliceosome B complex, DDX39B is responsible for regulating the recognition and pairing of U2 snRNP with the branch site during pre-mRNA splicing, ensuring accurate intron excision and exon ligation. Additionally, it plays a central role in mRNA nuclear export by interacting with the TREX (transcription-export) complex to transport mature mRNA from the nucleus to the cytoplasm for translation. In recent years, the functional scope of DDX39B has expanded to other biological areas, including participation in homologous recombination repair during DNA damage repair, as well as influencing cell proliferation and apoptosis through regulating the stability of specific mRNAs. Furthermore, DDX39B has been found to be associated with various diseases: it is overexpressed in multiple tumors (such as hepatocellular carcinoma and lung cancer), promoting tumor progression by enhancing the splicing and nuclear export of oncogenic mRNAs, and is correlated with poor patient prognosis. In viral infections, certain viruses hijack DDX39B to facilitate the processing and nuclear export of their own RNAs. Meanwhile, polymorphisms in the DDX39B gene have also been implicated in susceptibility to autoimmune diseases such as rheumatoid arthritis, highlighting the bridging role of this protein between fundamental RNA metabolism and disease development.
