Background
DCTD, also known as dCMP deaminase, is encoded by the human DCTD gene and is a homotypic hexameric enzyme that plays a critical role in pyrimidine nucleotide metabolism. This protein is approximately 178 amino acids in length with a molecular weight of about 20 kDa, is primarily localized in the cytoplasm, and its enzymatic activity depends on zinc ions as a cofactor. Functionally, DCTD catalyzes the hydrolytic deamination of deoxycytidylate (dCMP) to convert it into deoxyuridylate (dUMP), which is the direct substrate for thymidylate synthase in the synthesis of dTMP. Therefore, DCTD provides the essential precursors for DNA replication and repair through this key step. The activity of this enzyme is tightly regulated by allosteric mechanisms, with its activator dCTP enhancing its activity while the end product dTTP inhibits it, thereby precisely maintaining the balance of the intracellular deoxynucleotide pool. In addition, the DCTD protein undergoes various post-translational modifications, including phosphorylation, ubiquitination, and acetylation, which may participate in the fine-tuning of its activity and function.

IHC shows positive staining in paraffin-embedded human kidney. Anti-Dctd antibody was used at 1/200 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
