Background
Disabled-2 (DAB2) is a multifunctional adaptor protein involved in various cellular processes, including clathrin-mediated endocytosis, signal transduction, and regulation of key signaling pathways such as Wnt, MAPK, and TGF-β. Initially identified as a tumor suppressor in ovarian cancer, DAB2 is downregulated in several cancers, including breast, lung, and colorectal cancer, contributing to tumor progression. It contains functional domains like the phosphotyrosine-binding (PTB) domain and proline-rich SH3 domain, which enable it to interact with multiple proteins and regulate pathways involved in cell differentiation, proliferation, and epithelial-mesenchymal transition. DAB2 also plays a role in platelet activation and aggregation, with different isoforms expressed in human and mouse platelets. Additionally, it is essential for embryonic development and maintaining epithelial cell polarity.

IHC shows positive staining in paraffin-embedded human placenta. Anti-DAB2 antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
