Background

Cytochrome b (CYTB) is a crucial transmembrane protein encoded by the mitochondrial DNA in most eukaryotes, serving as the central catalytic subunit of Complex III (the cytochrome bc1 complex) within the electron transport chain. This protein contains two heme groups, heme bL and heme bH, which facilitate the transfer of electrons from ubiquinol to cytochrome c while simultaneously pumping protons across the inner mitochondrial membrane to generate the electrochemical gradient necessary for ATP synthesis via oxidative phosphorylation. Due to its essential role in cellular respiration, CYTB is highly conserved across species yet accumulates mutations at a rate suitable for phylogenetic studies, making it a standard molecular marker for evolutionary biology, species identification, and forensic analysis, while specific mutations in the human CYTB gene are also linked to mitochondrial diseases affecting energy-intensive tissues such as muscle and the nervous system.

Immunohistochemistry

Immunocytochemistry

ICC shows positive staining in C2C12 cells. Anti-CYTB antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
