Background
Cyclin E2, encoded by the CCNE2 gene, is a cell cycle protein that serves as an activating subunit of CDK2 and CDK3, playing a central regulatory role in the G1-to-S phase transition. Its expression levels and kinase activity peak at the G1/S boundary, driving the initiation of DNA replication through phosphorylation of substrates such as the Rb protein. Compared to its family member Cyclin E1, Cyclin E2 exhibits a unique high-expression pattern in normal tissues such as the testis, thymus, and brain, and is significantly upregulated in various tumor cells, where it has been closely linked to chemotherapy resistance and poor prognosis. As a short-lived protein, its stability is regulated by the ubiquitin-proteasome pathway and precisely controlled by phosphorylation-mediated degradation.

IHC shows positive staining in paraffin-embedded human tonsil. Anti-Cyclin E2 antibody was used at 1/100 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in HeLa cells. Anti- Cyclin E2 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
