Background

Cyclin B1 is a key regulatory protein that drives cells into mitosis. As the regulatory subunit of CDK1, it binds CDK1 to form the maturation-promoting factor (MPF), and activation of this complex is the central event initiating mitosis. Cyclin B1 protein levels begin to rise in S phase, peak at the G2/M transition, and are rapidly degraded during the metaphase-to-anaphase transition. Its subcellular localization is dynamic: during interphase it is mainly retained in the cytoplasm, and before mitotic onset, phosphorylation mediated by kinases such as PLK1 promotes its nuclear translocation, enabling it to phosphorylate mitotic substrates such as lamin and condensin. In tumor biology, Cyclin B1 is overexpressed in a variety of malignancies, including breast, colorectal, and prostate cancers, and is associated with tumor aggressiveness and poor prognosis; it is also recognized as one of the tumor antigens targeted by tumor immune surveillance.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human tonsil. Anti-Cyclin B1 antibody was used at 1/2000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

IHC shows positive staining in paraffin-embedded human colon.

Immunocytochemistry

ICC shows positive staining in HeLa cells. Anti-Cyclin B1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
