Background
CUX1 is a critically important transcription factor widely expressed in the human body. Its core function is to act as a "regulatory switch" for gene expression, playing a key role in development, cell differentiation, and cell cycle progression by binding to specific DNA sequences, and it typically serves as a transcriptional repressor of development-related genes. The CUX1 protein has a unique structure, containing three CUT domains and a homeodomain, which are the basis for its DNA-binding capacity. Its function exhibits "dosage sensitivity," meaning that even subtle changes in protein levels can have significant effects. In cancer research, CUX1 is considered a haploinsufficient tumor suppressor gene, where inactivation or mutation of one allele (commonly seen in various tumors such as acute myeloid leukemia) disrupts gene regulatory networks, thereby promoting tumorigenesis. Furthermore, CUX1's functions are not limited to the nucleus; it may also participate in Golgi vesicular transport, and its dysfunction has also been associated with diseases such as developmental delay.
