Background
Connective tissue growth factor (CTGF, also termed CCN2) is a 38 kDa, cysteine-rich, secreted matricellular protein of the CCN family that orchestrates cell adhesion, migration, proliferation, differentiation, angiogenesis, and extracellular matrix (ECM) remodeling by binding integrins, heparan sulfate proteoglycans, and growth factors such as TGF-β and VEGF through its four conserved domains (IGFBP, VWC, TSP-1, and cysteine-knot CT); physiologically it is indispensable for embryonic skeletal, pancreatic, ovarian, and vascular development, whereas dysregulated CTGF drives fibrosis, diabetic retinopathy, atherosclerosis, and tumorigenesis, making it a prominent therapeutic target now being pursued with monoclonal antibodies in pre-clinical and clinical trials.
