Background

CTBP1 (C-terminal binding protein 1) is a transcriptional corepressor that is highly conserved across metazoans, named for its initial discovery as a protein that binds to the C-terminal region of the adenoviral E1A oncoprotein. Structurally, CTBP1 shares extensive homology with the D-2-hydroxyacid dehydrogenase family and contains a conserved NAD(H)-binding domain and a Rossmann fold; however, dehydrogenase activity in vitro is not required for its transcriptional repression function—rather, the conformational changes induced by NAD(H) binding and oligomerization (especially tetramer formation) are key to its corepressor activity. CTBP1 primarily recognizes and binds the PXDLS motif present in many cellular transcription factors through a hydrophobic cleft at its N-terminus, and then acts as a scaffold to recruit chromatin-modifying enzymes such as histone deacetylases, methyltransferases, and demethylases to the promoters of target genes, repressing the expression of numerous targets, including genes involved in apoptosis, cell adhesion, and epithelial–mesenchymal transition. In addition, CTBP1 participates in extranuclear processes such as Golgi membrane fission and brown adipocyte differentiation, and its dysregulated expression is associated with poor prognosis in multiple cancers, making it a potential drug target.

Immunocytochemistry

ICC shows positive staining in SH-SY5Y cells. Anti-CTBP1 antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
