IHC shows positive staining in paraffin-embedded Rat hypothalamus. Anti-CRF antibody was used at 1/100 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Background
Corticotropin-releasing factor (CRF), also known as corticotropin-releasing hormone (CRH), is a 41-amino acid peptide hormone primarily synthesized in the paraventricular nucleus of the hypothalamus that serves as the principal neuroregulator of the hypothalamic-pituitary-adrenal (HPA) axis and the body's central stress response system. Upon release into the hypophyseal portal circulation, CRF binds with high affinity to G protein-coupled CRF receptors (specifically CRFR1 and CRFR2) on corticotroph cells in the anterior pituitary to stimulate the synthesis and secretion of adrenocorticotropic hormone (ACTH), which subsequently triggers glucocorticoid production in the adrenal cortex, while simultaneously acting as a neuromodulator within extrahypothalamic brain regions to orchestrate autonomic, behavioral, and immune adaptations to stressors such as anxiety, arousal, and appetite suppression. Dysregulation of CRF signaling has been extensively implicated in the pathophysiology of stress-related psychiatric disorders including major depression, post-traumatic stress disorder, and anxiety disorders, as well as metabolic and inflammatory conditions, making the CRF system a critical therapeutic target for modulating maladaptive stress responses and restoring neuroendocrine homeostasis.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
