Background

The CoREST protein (primarily encoded by the RCOR1 gene, also known as CoREST1) was initially identified as a key corepressor of the transcriptional repressor REST (RE1-Silencing Transcription Factor), functioning by binding to REST to silence neuron-specific gene expression in non-neuronal cells, thereby playing an important role in maintaining cellular identity. Structurally, the CoREST protein contains two SANT domains and an ELM2 domain, which serve as the fundamental basis for its role as a molecular scaffold in recruiting histone-modifying enzymes. Its most distinctive function is to act as the core scaffold of the LSD1-CoREST-HDAC complex, simultaneously binding the histone demethylase LSD1 and the histone deacetylases HDAC1/2, thereby integrating two distinct epigenetic modifying enzymes into a single complex to coordinately regulate chromatin status and gene transcription. Notably, the human genome contains three CoREST family members (CoREST1, CoREST2, and CoREST3), which exhibit differences in their binding affinities for LSD1 and HDAC as well as their transcriptional repressive activities—for instance, CoREST2 binds more weakly to HDAC1/2 due to specific amino acid variations, endowing different complexes with the capacity for fine-tuned regulation of gene expression. Furthermore, recent studies have revealed that CoREST proteins function far beyond the scope of "REST corepressors," as they can independently regulate biological processes such as neural development in a REST-independent manner and play important roles in various diseases.
