Background

CNBP (Cellular Nucleic Acid-Binding Protein), also known as ZNF9, is a highly conserved, single-stranded nucleic acid-binding protein that functions as a versatile nucleic acid chaperone. It plays a critical role in regulating both transcription and translation by remodeling nucleic acid secondary structures and modulating the activity of trans-acting factors. CNBP is essential for maintaining sustained immune responses (such as prolonged IL-6 expression) and proper embryonic development. Clinically, abnormal expansions of the CCTG repeat in the first intron of the CNBP gene are the direct cause of myotonic dystrophy type 2 (DM2), a multisystem disorder.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-CNBP antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
