Background

CLEC7A (C-type lectin domain family 7 member A) encodes the protein Dectin-1, a key member of the C-type lectin receptor family that functions as a pattern recognition receptor central to antifungal immunity. The gene is located on human chromosome 12p13.2 and is broadly expressed in tissues including bone marrow and appendix.

Structure and Isoforms: Dectin-1 is a type II transmembrane glycoprotein of approximately 28 kDa, containing an extracellular C-type lectin-like domain (CRD), a transmembrane region, and a cytoplasmic domain. The cytoplasmic tail harbors an immunoreceptor tyrosine-based activation motif (ITAM) essential for signal transduction. Alternative splicing generates at least eight isoforms (A-H), with isoform A representing the full-length functional form; isoform B lacks the stalk region, while others harbor deletions or insertions in the CRD or transmembrane domains.
