Background
CLEC5A, also known as C-type lectin domain family 5 member A, is a transmembrane receptor coupled to the adaptor protein DAP12 (DNAX-activating protein of 12 kDa) and belongs to the C-type lectin-like receptor superfamily. Its extracellular region contains a single C-type lectin-like domain (CTLD), which is connected via a transmembrane segment to the ITAM motif of intracellular DAP12, mediating downstream signal transduction. CLEC5A is predominantly expressed on myeloid immune cells, including monocytes/macrophages, dendritic cells, and neutrophils, serving as a critical pattern recognition receptor (PRR) for the recognition of pathogen-associated molecular patterns (PAMPs) and damage-associated molecular patterns (DAMPs). Functionally, CLEC5A participates in the regulation of inflammatory responses, macrophage polarization, and the tumor immune microenvironment, playing pivotal roles in sepsis, viral infections, and tumor progression. In recent years, CLEC5A has garnered significant attention as a novel immunotherapeutic target, with bispecific antibodies, antibody-drug conjugates (ADCs), and small-molecule inhibitors actively under development, demonstrating promising clinical application prospects in the treatment of solid tumors such as breast cancer and osteosarcoma, as well as sepsis-associated lung injury.
