Background

PARP (poly(ADP-ribose) polymerase) is a family of nuclear enzymes that detect and signal DNA strand breaks, catalyzing the transfer of ADP-ribose units from NAD to target proteins to create poly(ADP-ribose) chains that recruit DNA repair factors, thereby playing a central role in base-excision and single-strand break repair; pharmacological inhibition of PARP1/2 is synthetically lethal in BRCA1/2-mutated tumors because it traps the enzyme on DNA, preventing repair of replication-associated double-strand breaks that cannot be accurately rejoined in homologous-recombination–deficient cells, a strategy that underlies several approved cancer therapies.
