Background
Claudin-9 is a specialized integral membrane protein belonging to the claudin family, which serves as the primary structural and functional component of tight junctions in epithelial and endothelial cells, playing a critical role in regulating paracellular permeability and maintaining cell polarity. Specifically expressed in distinct tissues such as the inner ear (particularly within the organ of Corti), kidney, and certain regions of the gastrointestinal tract, Claudin-9 functions by forming charge-selective pores or seals between adjacent cells that control the passive diffusion of ions and small molecules; notably, mutations in the CLDN9 gene have been strongly linked to nonsyndromic hearing loss in humans, as the protein is essential for preserving the unique ionic composition of the endolymph required for proper auditory signal transduction, while its dysregulation or aberrant expression has also been observed in various carcinomas, suggesting a potential dual role in both physiological barrier maintenance and pathological processes like tumor progression or metastasis.
