Background
Claudin-9 is a 211-amino-acid tetraspan tight-junction protein that seals the paracellular space between epithelial and endothelial cells while selectively controlling ion and solute permeability; it is highly expressed in the inner ear, lung, testis and epidermis, where its absence or mutation causes deafness, skin barrier defects and increased susceptibility to viral entry, and its transcription is driven by grainyhead-like transcription factors that bind a conserved 5′-enhancer, making claudin-9 a critical determinant of tissue-specific barrier integrity, ion homeostasis and innate defense.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
