Background

CHX10, also known as VSX2 (Visual System Homeobox 2), is a paired-like homeodomain transcription factor that serves as a master regulator in vertebrate eye development, playing indispensable roles in the specification, proliferation, and differentiation of retinal progenitor cells into various neuronal lineages, particularly bipolar cells and Müller glia. Expressed primarily in the developing neural retina, CHX10 functions by binding to specific DNA sequences to activate or repress target genes involved in cell cycle control and fate determination, thereby ensuring the proper formation of retinal layers and overall eye morphology. Mutations in the CHX10 gene are associated with severe congenital eye disorders in both humans and animals, such as microphthalmia (abnormally small eyes) and anophthalmia (absence of eyes), highlighting its critical importance in oculogenesis; furthermore, its conserved expression pattern across species underscores its fundamental evolutionary role in establishing the visual system, making it a key marker for studying retinal development and a potential target for regenerative therapies aimed at restoring vision through stem cell-derived retinal tissues.

IHC shows positive staining in paraffin-embedded human retina. Anti-CHX10 antibody was used at 1/500 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
