Background

CHMP1a (Charged Multivesicular Body Protein 1a) is a protein encoded by the CHMP1A gene in humans and belongs to the ESCRT-III (Endosomal Sorting Complex Required for Transport) family. It plays a crucial role in the formation and sorting of multivesicular bodies (MVBs), which are involved in the degradation of proteins in lysosomes. CHMP1a is also known as Chromatin Modifying Protein 1A and has been identified as a novel tumor suppressor gene, particularly in pancreatic and embryonic kidney tumor cells. It can inhibit cell proliferation and tumor growth by regulating the expression of proteins like p53. Additionally, CHMP1a interacts with other proteins such as SKD1 and LIP5, which are involved in the endosomal sorting process.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human testis. Anti-CHMP1a antibody was used at 1/50 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in 293T cells. Anti- CHMP1a antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
