Background

CHCHD10 (Coiled-coil-helix-coiled-coil-helix domain containing 10) is a small, nuclear-encoded protein primarily localized to the mitochondrial intermembrane space, where it plays a critical structural and functional role in maintaining mitochondrial cristae integrity and facilitating proper oxidative phosphorylation. As a member of the CHCH domain-containing protein family, CHCHD10 forms disulfide-bonded homodimers or heterodimers with its paralog CHCHD2, interacting with key components of the mitochondrial contact site and cristae organizing system (MICOS), particularly mitofilin (IMMT), to stabilize the inner mitochondrial membrane architecture. Beyond its structural duties, CHCHD10 is implicated in regulating mitochondrial dynamics, apoptosis, and cellular stress responses, and mutations in the CHCHD10 gene have been strongly associated with a spectrum of neurodegenerative disorders, including amyotrophic lateral sclerosis (ALS), frontotemporal dementia (FTD), and mitochondrial myopathy, highlighting its essential contribution to neuronal health and mitochondrial homeostasis.
