Background
CEP55 (Centrosomal Protein 55), also known as FLJ10540 or C5orf38, is a coiled-coil protein primarily localized to the midbody during the final stages of cell division, where it plays a critical role in cytokinesis and abscission by recruiting essential machinery such as the ESCRT-III complex and ALIX to ensure the proper physical separation of daughter cells. Beyond its canonical function in mitosis, CEP55 is significantly overexpressed in various malignant tumors, including breast, lung, and liver cancers, acting as an oncogene that promotes cell proliferation, migration, and invasion while inhibiting apoptosis, often through interactions with key signaling pathways like PI3K/AKT and MAPK. Due to its restricted expression in normal adult tissues—being largely absent except in testis—and its high prevalence in cancer cells, CEP55 has emerged as a promising tumor-specific antigen for immunotherapy and a potential prognostic biomarker, linking defective cytokinesis directly to genomic instability and tumorigenesis.
