IHC shows positive staining in paraffin-embedded human colon. Anti-CENP-F antibody was used at 1/500 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Background
CENP-F (Centromere Protein F), also known as mitosin, is a large, transient kinetochore protein that plays a critical role in ensuring accurate chromosome segregation during cell division. Primarily localized to the kinetochores from late G2 phase through metaphase, CENP-F functions as a key scaffold and regulatory hub that facilitates the attachment of spindle microtubules to chromosomes. It interacts with various components of the mitotic machinery, including dynein and the nuclear pore complex protein Nup133, to coordinate spindle assembly checkpoint signaling and maintain genomic stability. Due to its essential function in mitosis and its frequent overexpression in various malignancies, CENP-F is often studied as a potential biomarker for cancer prognosis and a target for anti-mitotic therapies.
