1. Reagent preparation steps: Cell working solution (1X): Cell recovery solution (10X) was diluted into cell working solution (1X) using 4℃ PBS (1X). 2. Hydrogel dissolution step: All steps were performed in a sterile operating bench, and the operating steps were as follows: 1. After the 3D cell culture is formed, carefully aspirate the culture medium to remove it and wash it with PBS (1X). The PBS was removed by pipetting, and 1 mL of pre-cooled cell working solution (1X) was added to cover the droplets and reacted at room temperature for 5 min. 2. Gently blow with a pipette until the hydrogel is completely dissolved. 3. The solution containing the cell spheres was drawn into a centrifuge tube, centrifuged at 250g for 5 minutes, the supernatant liquid was removed, and the pelleted cell spheres were collected for subsequent experimental analysis. |
| Description | The cell recovery solution can recover cells cultured in a hydrogel for downstream protein analysis. This solution does not contain any enzyme components, can dissolve the hydrogel without affecting cell activity, does not require enzyme digestion or long-term high-temperature incubation, and can quickly and efficiently recover organoids/cells from the hydrogel system within 5 minutes. |
| Storage Temp. | Store at 2-8°C with a shelf life of 12 months. |
| Applications | The hydrogel was dissolved and the cells were recovered. |
