Background
CDP-diacylglycerol synthase 2 (CDS2), also known as CDP-DAG synthase 2, is a critical membrane-bound enzyme located primarily in the endoplasmic reticulum that catalyzes the conversion of phosphatidic acid and cytidine triphosphate (CTP) into cytidine diphosphate-diacylglycerol (CDP-DAG), serving as the committed and rate-limiting step in the de novo biosynthesis of major phospholipids such as phosphatidylinositol, phosphatidylglycerol, and cardiolipin. As one of two isoforms in mammals (the other being CDS1), CDS2 plays an indispensable role in maintaining cellular membrane integrity, facilitating intracellular signaling pathways dependent on phosphoinositides, and regulating lipid homeostasis, with its expression and activity being tightly linked to metabolic processes where dysregulation has been associated with various pathologies including lipodystrophy, insulin resistance, and altered susceptibility to viral infections due to its involvement in modifying host membrane composition required for viral replication.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human liver. Anti-CDS2 antibody was used at 1/250 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
