Background

CDP-diacylglycerol synthase 1 (CDS1), also known as CDP-DAG synthase 1, is a critical integral membrane enzyme primarily located in the endoplasmic reticulum that catalyzes the conversion of phosphatidic acid (PA) and cytidine triphosphate (CTP) into cytidine diphosphate-diacylglycerol (CDP-DAG), serving as the committed and rate-limiting step in the de novo biosynthesis of major phospholipids such as phosphatidylinositol (PI), phosphatidylglycerol (PG), and cardiolipin. As a central hub in lipid metabolism, CDS1 regulates the flux of PA toward phospholipid production rather than triglyceride synthesis or signaling pathways, thereby maintaining cellular membrane integrity, facilitating signal transduction cascades like the PI3K/Akt pathway, and supporting mitochondrial function through cardiolipin generation; its activity is tightly regulated by feedback inhibition from downstream products and transcriptional mechanisms, and dysregulation or mutations in the CDS1 gene have been implicated in severe metabolic disorders, including congenital lipodystrophy, insulin resistance, and altered susceptibility to viral infections due to compromised membrane architecture.
