Background

CDK9 (cyclin-dependent kinase 9) is the catalytic subunit of the positive transcription elongation factor b (P-TEFb), which phosphorylates the RNA polymerase II C-terminal domain and negative elongation factors to promote productive elongation of RNA synthesis; in its activated form CDK9 is complexed with cyclin T or cyclin K and is essential for expression of most RNA polymerase II-dependent genes, and because it controls transcriptional elongation of key regulators of cell proliferation and survival—especially MCL-1, MYC and cyclin D1—CDK9 is an attractive therapeutic target in cancer, inflammation and viral infections, with several ATP-competitive inhibitors such as flavopiridol, dinaciclib and alvocidib already in clinical trials.
