Background
Cyclin-dependent kinase 6 (CDK6) is a serine/threonine protein kinase that plays a pivotal role in regulating the cell cycle, specifically during the G1 to S phase transition, by forming active heterodimeric complexes with D-type cyclins (Cyclin D1, D2, or D3). Upon binding to these cyclins, the CDK6 complex phosphorylates the retinoblastoma protein (Rb), which leads to the release of E2F transcription factors and subsequently drives the expression of genes required for DNA synthesis and cell proliferation. Beyond its canonical function in cell cycle control, CDK6 is implicated in various physiological processes including hematopoiesis, differentiation, and metabolism, and its dysregulation—often through gene amplification or overexpression—is frequently observed in numerous cancers, such as breast cancer, lymphoma, and glioblastoma, making it a significant therapeutic target for selective CDK4/6 inhibitors like palbociclib, ribociclib, and abemaciclib, which are widely used in clinical settings to halt tumor growth by inducing G1 cell cycle arrest.

Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
