Background

CDH17 (Cadherin-17, also known as LI-cadherin or HPT-1) is a unique member of the 7D-cadherin subclass within the cadherin superfamily. Structurally, it features an extracellular region containing seven cadherin repeats and a very short cytoplasmic domain (only 18–22 amino acids), which prevents it from binding to catenins like classical cadherins. Under normal physiological conditions, CDH17 is mainly expressed in the embryonic liver and the adult small intestine and colon, where it functions as a cell adhesion molecule and peptide transporter. However, it is aberrantly overexpressed in various gastrointestinal malignancies, including hepatocellular carcinoma, gastric cancer, colorectal cancer, and pancreatic cancer, and is closely associated with tumor invasion, metastasis, and poor prognosis. Studies have shown that CDH17 promotes tumor cell proliferation, survival, migration, and invasion by activating key signaling pathways such as Wnt, NF-κB/MAPK, and Akt. Due to its highly specific expression pattern in advanced gastrointestinal cancers, CDH17 has been recognized as a promising novel diagnostic biomarker and a highly potential target for cancer therapy.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human colon. Anti-CDH17 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
