Background
Mouse CD97 (mCD97) is a member of the adhesion G protein-coupled receptor (aGPCR) family. Its extracellular region undergoes alternative splicing to generate isoforms containing different numbers of EGF-like domains and exists in a glycosylated form. Its N-terminal domain binds to the ligand CD55 (decay-accelerating factor), while the transmembrane region mediates signal transduction. Functionally, CD97 acts as a mechanosensor that regulates the localization of splenic marginal zone B cells and T cell-independent antibody production by sensing blood flow shear stress, and also maintains dendritic cell homeostasis. Additionally, it participates in granulocyte migration, hematopoietic stem cell differentiation, and tumor microenvironment remodeling. Clinically, CD97 is highly expressed in malignant tumors such as gastric cancer, where its small isoform promotes tumor invasion and metastasis, serving as both a prognostic marker and a potential therapeutic target. Moreover, its immunomodulatory roles in inflammatory diseases offer new avenues for intervention strategies.
