Background
CD94, encoded by KLRD1, is a type II transmembrane C-type lectin glycoprotein expressed predominantly on natural killer (NK) cells and subsets of CD8⁺ T cells, where it covalently assembles with NKG2A, C, E or F subunits to form disulfide-linked heterodimers that recognize the non-classical MHC-I molecule HLA-E (or Qa-1 in mice) and deliver either inhibitory or activating signals that finely tune cytotoxicity, cytokine secretion and survival of effector lymphocytes, thereby playing a central role in innate and adaptive immunity, tumor immune evasion, viral defense and pregnancy tolerance.

Immunocytochemistry
ICC shows positive staining in NK-92 cells (top panel) and negative staining in Jurkat cells (below panel). Anti-CD94 antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
