Background
CD70, also known as CD27 ligand, is a type Ⅱ transmembrane protein that plays a crucial role in the immune system. It is primarily expressed on antigen - presenting cells (APCs) such as dendritic cells, macrophages, and B cells, as well as on activated T cells. CD70 interacts with its receptor CD27, which is mainly found on T cells, B cells, and natural killer (NK) cells. This interaction provides important co - stimulatory signals that promote T cell activation, proliferation, and differentiation, and also enhances B cell and NK cell functions. Additionally, CD70 is involved in the regulation of immune responses, including the maintenance of immune homeostasis and the induction of immune - mediated tumor cell killing. Abnormal expression of CD70 has been associated with various diseases, such as autoimmune disorders and certain types of cancers. Therefore, CD70 is considered a promising target for the development of immunotherapies.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in U266B1 cells (top panel) and negative staining in Jurkat cells (below panel). Anti-CD70 antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
