Background
CD68 is a heavily glycosylated, 110 kDa type I transmembrane glycoprotein of the lysosome-associated membrane protein (LAMP) family (also called LAMP-4) that is highly expressed on human monocytes, macrophages, osteoclasts, and other myeloid cells, where it predominantly localizes to lysosomes and endosomes and participates in phagocytosis, debris clearance, and inflammatory responses via its scavenger-receptor activity, while alternative splicing produces multiple isoforms and its presence on tumor cells and ability to bind selectins implicate it in metastasis, making it a widely used histological macrophage marker.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded mouse spleen. Anti-CD68 antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
