Background
CD44 variable alternative exon 6 (CD44 V6) is a variant isoform of the CD44 gene resulting from alternative splicing of RNA. CD44 is a cell adhesion molecule that is present in various cell types and plays crucial roles in biological processes such as cell migration, proliferation, and differentiation. The CD44 V6 isoform specifically, has been implicated in tumor development and metastasis. Research has shown that CD44v6 is expressed by all colorectal cancer stem cells (CR-CSCs). The expression of CD44v6 is essential for the migration and formation of metastatic tumors, achieved through the activation of the WNT-β-catenin signaling pathway. Propelling evidence recommends that CD44v6 compared to CD44 has a more specific expression in CSCs and may have a higher diagnostic value.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human tonsil. Anti- CD44v6 antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
IHC shows positive staining in paraffin-embedded human prostate.
Negative control: IHC shows negative staining in paraffin-embedded human spleen.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
Immunocytochemistry
ICC shows positive staining in A431 cells (top panel) and negative staining in Daudi cells (below panel). Anti-CD44v6 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
