Principle of Separation

CD4⁺ cells are magnetically labeled using CD4 Nanobeads for humans. The cell suspension is then loaded into a separation column placed within the sorter's magnetic field. The magnetically labeled CD4⁺ cells remain retained in the column, while the unlabeled cells flow through; this fraction constitutes the negative fraction after removal of CD4⁺ cells. After removing the separation column from the magnetic field, the magnetically captured CD4⁺ cells can be eluted, yielding the positive sorted fraction. If higher purity is required, the positive sorted fraction containing CD4⁺ cells must be further separated using a second column.

Background

CD4 NanoBeads are designed to isolate human cells based on the expression of the CD4 antigen. CD4 is a 55 kDa protein that is highly expressed on helper T cells in the thymus and peripheral blood. In helper T cells, CD4 functions as a co-molecule, assisting the TCR heterodimer in recognizing MHC class II–peptide complexes. CD4 is also expressed on monocytes and dendritic cells, though at lower levels than in CD4+ helper T cells. When bound to certain chemokine receptors, CD4 serves as the primary high-affinity receptor for HIV.

Validation Data

Enrich or deplete CD4+ T cells from human PBMCs. Stain CD4+ cells with anti-CD4 (clone: RPA-T4) Brilliant Violet 421™, and gate on the live cell population for analysis.
