Background
CD3γ (Cluster of Differentiation 3 gamma) is a critical subunit of the T cell receptor (TCR) complex. It is a type I transmembrane glycoprotein belonging to the immunoglobulin superfamily. The CD3γ chain, together with CD3δ, CD3ε, and the CD3ζ homodimer, forms the invariant CD3 signaling module that associates with the variable TCRαβ or TCRγδ chains.
The primary function of CD3γ is to mediate signal transduction upon antigen recognition. Like other CD3 subunits, CD3γ contains three immunoreceptor tyrosine-based activation motifs (ITAMs) in its cytoplasmic domain. Upon TCR engagement with peptide-MHC complexes, these ITAMs are phosphorylated by Src-family kinases (LCK and FYN), initiating downstream signaling cascades that lead to T cell activation, proliferation, and differentiation.
