Background

CD177 (also known as NB1 glycoprotein or HNA-2a antigen) is a glycosylphosphatidylinositol (GPI)-anchored glycoprotein of approximately 50–60 kDa belonging to the Ly-6/uPAR superfamily, expressed mainly on the plasma membrane and secondary granules of neutrophils, and present only on a subset of about 45%–65% of neutrophils, with the expressing proportion varying among healthy individuals. Its classic function is to serve as a high-affinity receptor for proteinase 3 (PR3) on the neutrophil surface, forming the CD177–PR3 membrane complex that maintains high-density presentation of PR3 on the cell surface; this complex constitutes a key target for autoantibody attack in PR3-ANCA-associated vasculitis, and blocking the binding of CD177 to PR3 can significantly reduce ANCA-induced neutrophil respiratory burst. In addition, CD177 can participate in neutrophil transendothelial migration by binding PECAM-1 on endothelial cells, and may influence the balance between adhesion and migration by regulating β2 integrin signaling. Recent studies have further revealed that CD177 is also expressed on the surface of some tumor-infiltrating regulatory T cells and is associated with enhanced immunosuppressive activity and tumor immune evasion. Clinically, absence of CD177 expression is associated with the HNA-2-negative phenotype and can lead to transfusion-related acute lung injury and neonatal alloimmune neutropenia, while changes in the proportion of CD177-expressing neutrophils are also regarded as a potential biomarker for diseases such as myeloproliferative neoplasms and inflammatory bowel disease.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human colon. Anti-CD177 antibody was used at 1/2000dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
