Background
CD151, also known as tetraspanin-24 or PETA-3, is a widely expressed transmembrane glycoprotein belonging to the tetraspanin superfamily that plays a pivotal role in regulating cell adhesion, migration, and signaling by organizing multimolecular complexes within the plasma membrane. Functioning primarily as a molecular organizer, CD151 forms stable associations with integrins, particularly laminin-binding integrins such as α3β1, α6β1, and α6β4, thereby strengthening cell-matrix interactions and facilitating the assembly of specialized microdomains often referred to as "tetraspanin webs" that recruit various signaling molecules, growth factor receptors, and proteases. This protein is critically involved in diverse physiological processes including embryonic development, angiogenesis, wound healing, and platelet function, while its dysregulation is strongly implicated in pathological conditions, most notably cancer progression, where elevated CD151 expression correlates with enhanced tumor cell motility, invasion, metastasis, and poor prognosis in carcinomas of the breast, lung, liver, and pancreas by modulating integrin-dependent signaling pathways like PI3K/Akt and MAPK/ERK; furthermore, CD151 serves as an entry cofactor for certain viruses, such as hepatitis C virus and human papillomavirus, making it a significant target for therapeutic interventions aimed at inhibiting metastatic spread or viral infection.
