The single-cell suspension was subjected to CD138⁺ cell depletion using CD138 Nanobeads, S or L Separation Columns, and a magnetic sorter. Subsequently, fluorescent staining was performed with PE Mouse Anti-Human CD138 Antibody (S-R553) (STD Cat: S0B5646), and the live cell population was gated for analysis.

| Principle of Separation | CD138⁺ cells are magnetically labeled using CD138 Nanobeads. The cell suspension is then loaded into a separation column placed within the sorter's magnetic field. The magnetically labeled CD138⁺ cells remain retained in the column, while the unlabeled cells elute; this fraction constitutes the negative fraction after removal of CD138⁺ cells. After removing the separation column from the magnetic field, the magnetically captured CD138⁺ cells can be eluted, yielding the positive sorted fraction. If higher purity is required, the positive sorted fraction containing CD138⁺ cells must be further separated through a second column. |

Background
CD138 NanoBeads are specifically developed for the isolation of plasma cells from human peripheral blood mononuclear cells, bone marrow, or lymphoid tissues. CD138, also known as syndecan-1, is expressed on the surface of both normal and malignant human plasma cells and serves as a universal marker for identifying plasma cells. CD138 is also expressed on the basolateral membrane of endothelial cells, embryonic mesenchymal cells, vascular smooth muscle cells, and nerve cells. In mice, in addition to its expression on plasma cells, CD138 is also found on B-cell precursor cells in the bone marrow.
