Background

CCR5 (C-C chemokine receptor type 5) is a G protein-coupled receptor primarily expressed on the surface of immune cells, including T cells, macrophages, and dendritic cells, where it plays a pivotal role in mediating inflammatory responses by binding to specific chemokines such as CCL3, CCL4, and CCL5 to regulate leukocyte trafficking and activation. Beyond its physiological function in immunity, CCR5 gained significant scientific prominence as a major co-receptor, alongside CD4, for the entry of R5-tropic strains of Human Immunodeficiency Virus (HIV-1) into host cells, making it a critical target for antiretroviral therapies and gene-editing strategies aimed at conferring resistance to HIV infection, notably illustrated by the rare natural mutation CCR5-Δ32 which prevents viral entry and has been leveraged in groundbreaking curative approaches like stem cell transplants.
