Background
CCDC47 (Coiled-Coil Domain Containing Protein 47) is a multifunctional transmembrane protein primarily localized to the endoplasmic reticulum, playing a critical role in cellular calcium homeostasis regulation, protein processing, and the unfolded protein response. This protein is characterized by an N-terminal signal peptide, a central region containing multiple predicted coiled-coil domains that mediate protein-protein interactions, and a C-terminal transmembrane domain that anchors it to the endoplasmic reticulum membrane. As an important member of endoplasmic reticulum calcium-binding proteins, CCDC47 binds calcium ions and participates in regulating calcium store homeostasis within the ER lumen, thereby influencing calcium-dependent signaling and apoptotic processes. Additionally, this protein is involved in ER-associated protein degradation pathways and the formation of ER-mitochondria contact sites, helping to maintain functional coupling between organelles. Genetic studies have revealed that mutations in the CCDC47 gene are directly associated with a rare progressive neurological disorder—early-onset spastic paraplegia with optic atrophy and cognitive impairment (SPOAN syndrome)—highlighting the irreplaceable protective role of this protein in neurodevelopment and myelination processes.
