Background
CBX2 (Chromobox Protein Homolog 2) is a critical component of the Polycomb Repressive Complex 1 (PRC1) that functions as an epigenetic regulator by specifically recognizing and binding to histone H3 lysine 27 trimethylation (H3K27me3) marks deposited by PRC2, thereby facilitating chromatin compaction and the stable transcriptional silencing of target genes essential for embryonic development, stem cell pluripotency, and cellular differentiation. Unlike some other CBX family members, CBX2 possesses a unique AT-hook-like domain that enables direct DNA binding independent of histone modifications, allowing it to recruit PRC1 to specific genomic loci and contribute to higher-order chromatin architecture through phase separation mechanisms. Its dysregulation is frequently implicated in various malignancies, where aberrant CBX2 expression can drive tumorigenesis by repressing tumor suppressor genes or maintaining cancer stem cell states, making it a significant biomarker and potential therapeutic target in oncology and developmental biology research.
