Background

Cathepsin D is an aspartic endo-protease that is ubiquitously distributed in lysosomes. The main function of cathepsin D is to degrade proteins and activate precursors of bioactive proteins in pre-lysosomal compartments. Over-expression of cathepsin D stimulates tumorigenicity and metastasis as well as initiation of tumor apoptosis. This protease has been regarded an independent marker of poor prognosis in breast cancer being correlated with the incidence of clinical metastasis.

Flow cytometric analysis of 80% methanol fixed 0.1% Tween-20 permeabilized HepG2 (Human hepatocellular carcinoma epithelial cell) cells labelling Cathepsin D antibody at 1/500 dilution (0.1 μg) / (red) compared with a Rabbit monoclonal IgG isotype control (Black) and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (Blue). Goat Anti - Rabbit IgG Alexa Fluor 488 was used as the secondary antibody.

Immunocytochemistry

ICC shows positive staining in HepG2 cells. Anti- Cathepsin D antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue).
