Background
Caspase-12 is a cysteine protease specifically localized to the endoplasmic reticulum (ER) that plays a pivotal role in mediating ER stress-induced apoptosis triggered by stimuli such as calcium overload, hypoxia, and glucose deprivation. It is also closely associated with neurodegenerative processes, as it co-localizes with Alzheimer's disease-related proteins and mediates amyloid-beta-induced cytotoxicity. Notably, while Caspase-12 is fully functional in rodents, the human CASP12 gene contains a premature stop codon in most populations, rendering it a pseudogene with no protease activity.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded mouse kidney. Anti- Caspase-12 antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
