Background
Caspase-11 protein is an important inflammation-related cysteine-aspartic protease in mice, serving as a core component of the non-canonical inflammasome pathway and playing a key regulatory role in host defense and inflammatory responses. Unlike classical caspase-1, Caspase-11 can be directly recognized and bound by intracellular lipopolysaccharide (LPS, a major component of the Gram-negative bacterial cell wall), and this binding triggers its oligomerization and activation. Activated Caspase-11 cleaves and activates Gasdermin D (GSDMD) on one hand, which then forms pores in the cell membrane, inducing a pro-inflammatory form of cell death called pyroptosis, thereby exposing intracellular pathogens to the immune system. On the other hand, Caspase-11 can also activate the canonical NLRP3 inflammasome, indirectly promoting caspase-1 activation and the maturation and secretion of pro-inflammatory cytokines such as IL-1β and IL-18. Notably, the human homologs of Caspase-11 are Caspase-4 and Caspase-5, which possess similar LPS-sensing functions. Hyperactivation of Caspase-11 is closely associated with various inflammatory diseases, such as sepsis, endotoxic shock, and inflammatory bowel disease.
