Background

CAPRIN1 (Cell Cycle Associated Protein 1) is a multifunctional RNA-binding protein that plays critical roles in regulating cell proliferation, stress response, and innate immunity by modulating mRNA stability, translation, and signal transduction pathways. Primarily localized in cytoplasmic ribonucleoprotein granules known as stress granules, CAPRIN1 facilitates the assembly of these membraneless organelles under cellular stress conditions to temporarily repress translation and protect mRNAs from degradation, while also interacting with key signaling molecules like G3BP1 to influence cell cycle progression and apoptosis. Beyond its role in stress adaptation, CAPRIN1 has been implicated in tumorigenesis and viral defense mechanisms, as it can either promote or suppress cancer cell growth depending on context and serves as a regulator of antiviral responses by interacting with proteins involved in interferon signaling, making it a significant biomarker and potential therapeutic target in oncology and infectious disease research.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human breast cancer. Anti-CAPRIN1 antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in HeLa cells. Anti-CAPRIN1 antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Mouse IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).

ICC shows positive staining in NIH/3T3 cells. Anti-CAPRIN1 antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Mouse IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100.
