Background

Caprin-1 (Cytoplasmic Activation/Proliferation-Associated Protein 1) is an evolutionarily conserved RNA-binding protein whose N-terminal dimerization domain mediates homodimerization and assembly of ribonucleoprotein complexes, aggregating in cytoplasmic stress granules under cellular stress. Caprin-1 promotes cell proliferation by regulating mRNA translation and cell cycle progression, and its absence causes severe proliferative defects. It also acts as a Cyr61-interacting protein to drive osteosarcoma growth and lung metastasis, and is targeted by microRNAs such as miR-223 and miR-181a, contributing to tumor progression, invasion, and autophagy regulation in multiple malignancies including breast, gastric, and pancreatic cancers. Membrane-expressed CAPRIN-1 has been identified as a novel universal cancer therapeutic target; the humanized IgG1 monoclonal antibody TRK-950 targeting this protein has completed preclinical validation and entered Phase I clinical trials for advanced solid tumors, establishing Caprin-1 as a critical biomarker and therapeutic target bridging tumor molecular biology and precision medicine translational research.
