Background
CAP1, also known as Adenylate Cyclase-Associated Protein 1, is a highly conserved and multifunctional actin-regulatory protein that plays a pivotal role in cytoskeletal dynamics by coordinating actin monomer recycling and filament turnover. Structurally, CAP1 functions as a hexameric complex that bridges the activities of ADF/cofilin and profilin; it accelerates the dissociation of cofilin from ADP-actin monomers, facilitates the exchange of ADP for ATP on these monomers, and subsequently delivers the ATP-actin to profilin for efficient re-polymerization at the barbed ends of filaments. Beyond its canonical function in maintaining the global pool of polymerization-competent actin, CAP1 is implicated in various cellular processes including cell migration, endocytosis, and signal transduction pathways involving cyclic AMP, with dysregulation or mutations in the CAP1 gene being associated with developmental defects and immune disorders in humans, thereby highlighting its essential contribution to cellular architecture and physiological homeostasis.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human testis. Anti-CAP1 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
