Background
Calreticulin (CRT) is a highly conserved, multifunctional calcium-binding chaperone protein primarily residing in the lumen of the endoplasmic reticulumER), where it plays a pivotal role in maintaining cellular calcium homeostasis and facilitating the proper folding and quality control of newly synthesized glycopptides through its interaction with the calnexin/calreticulin cycle. Beyond its canonical intracellular functions, CRT exhibits significant "moonlighting" properties by translocating to the cell surface under stress conditions, such as immunogenic cell death, where it acts as a critical "eat-me" signal that promotes the phagocytosis of apoptotic or dying cells by dendritic cells and macrophages via binding to the LDL receptor-related protein 1 (LRP1/CD91), thereby bridging innate and adaptive immunity. Furthermore, extracellular CRT has been implicated in various pathological processes, including wound healing, angiogenesis, and autoimmune diseases like systemic lupus erythematosus, making it a subject of intense research for potential therapeutic interventions in cancer immunotherapy and inflammatory disorders.

Immunocytochemistry
ICC shows positive staining in NIH/3T3 cells. Anti-Calreticulin antibody was used at 1/250 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
