Background

Calpain 1 (CAPN1), also known as μ-calpain, is a ubiquitously expressed, calcium-dependent non-lysosomal cysteine protease in humans. It typically exists as a heterodimer consisting of an 80 kDa catalytic large subunit encoded by the CAPN1 gene and a 28 kDa regulatory small subunit encoded by the CAPNS1 gene. Calpain 1 is uniquely activated by micromolar levels of calcium ions, hence its name μ-calpain (micro-calpain), distinguishing it from its homolog, calpain 2, which requires millimolar calcium concentrations for activation. Under physiological conditions, Calpain 1 is localized to various cellular compartments, including the cytoplasm and mitochondria. It performs limited proteolytic cleavage of specific substrates and participates in key processes such as cytoskeletal remodeling, signal transduction, and cell migration. However, under pathological conditions such as ischemia-reperfusion injury and Alzheimer's disease, disruption of intracellular calcium homeostasis can lead to excessive activation of Calpain 1, resulting in aberrant cleavage of various functional proteins (e.g., apoptosis-inducing factor AIF and junctophilin-2 in the heart), thereby mediating cellular damage and dysfunction. Therefore, precise regulation of Calpain 1 is essential for maintaining cellular health and function, making it a potential therapeutic target for various diseases.
