Background
CALHM2 (Calcium Homeostasis Modulator 2) is a 323-amino-acid, four-pass transmembrane protein that oligomerizes into an undecameric, voltage-gated channel which—in addition to its originally described roles in taste-bud ATP release and neuronal excitability—has recently been shown to reside in the inner mitochondrial membrane where it functions as a Ca²⁺-inhibited import channel that translocates the ECHA subunit of the mitochondrial trifunctional protein into the matrix to enable fatty-acid β-oxidation, cristae integrity and normal respiratory metabolism, thereby coupling ion-channel gating to cellular energy supply and implicating CALHM2 in metabolic and neuropsychiatric disorders.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human stomach. Anti-CALHM2 antibody was used at 1/50 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in U2-OS cells. Anti-CALHM2 antibody was used at 1/50 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
